Expression of Genetically Controlled Resistance to Tobacco Mosaic Virus Infection in Isolated Tomato Leaf Mesophyll Protoplasts

چکیده
Phenotypic expression of different genes controlling resistance to tobacco mosaic virus (TMV) in tomato was analysed in the protoplast system using otherwise isogenic breeding lines. Genes Tm-2 and Tin-22 were not expressed and did not prevent TMV-L, a common tomato strain of TMV, infecting and multiplying. By contrast, homozygous gene Tin-I was able to express its effect in protoplasts as well as in leaf discs; no virus progeny were detected by fluorescent antibody staining or by infectivity assay up to 3 days after inoculation with TMV-L. Protoplasts and leaf discs homozygous for Tin-t, however, became infected with TMV-CH2, a tomato strain able to overcome the effects of Trn-~ in intact plants. Protoplasts from Lycopersicon peruvianum P. I. 128650 , known to have a high level of resistance to TMV, were as readily infected with TMV-L, and synthesized progeny virus as rapidly as protoplasts from susceptible tomato. This genotype seems to have no resistance expressible in isolated protoplasts.

Synthesis and localization of pathogenesis-related proteins in tobacco.
The PR1 family of pathogenesis-related proteins from tobacco (Nicotiana tabacum L.) leaves is induced by a variety of pathogenic and chemical agents and is associated with resistance to tobacco mosaic virus. The majority of the PR1 proteins did not copurify with mesophyll protoplasts (the major cell type of the leaf) isolated from tobacco mosaic virus-infected N. tabacum cv. Xanthi-nc leaves. H...

Protoplasts isolated from tobacco leaves were inoculated with cucumber mosaic virus (CMV) using a method slightly modified from that developed for inoculating tobacco mosaic virus. Multiplication of CMV was demonstrated by staining the protoplasm with fluorescent CMV-antibody, by the electron microscopy of ultrathin sections, and by assaying the infectivity in protoplasts. Seventy to 90% of pro...
متن کاملInfection of tobacco mesophyll protoplasts by tobacco mosaic virus.
It was proved that the protoplasts prepared from mesophyll of Nicotiana tabacum are infected by tobacco mosaic virus. The infection occurred when purified tobacco mosaic virus particles were added to a protoplast suspension in the presence of poly-L-ornithine. The virus multiplied in these protoplasts to a level of 10(6) virus particles per infected protoplast during 24 hours of incubation. The...

Tobacco mosaic virus (TMV) and Cucumber mosaic virus expressing green fluorescent protein (GFP) were used to probe the effects of salicylic acid (SA) on the cell biology of viral infection. Treatment of tobacco with SA restricted TMV.GFP to single-epidermal cell infection sites for at least 6 d post inoculation but did not affect infection sites of Cucumber mosaic virus expressing GFP. Microinj...
متن کاملDNA Synthesis in Tobacco Mosaic Virus-Infected Nicotiana tabacum Protoplasts and Regeneration of Persistently Infected Calli.
Tobacco mosaic virus infection of Nicotiana tabacum mesophyll protoplasts did not affect the pattern of chloroplast or total cellular DNA synthesis for at least 120 h when compared with that of mock-infected cells. Calli derived from infected protoplasts often showed large amounts of tobacco mosaic virus RNA and coat protein.
